Because of its structural similarity to heparin and heparan sulfate—biologically important molecules involved in blood anticoagulation, bacterial and viral infection, and other physiological and pathophysiological events—K5 polysaccharide offers a promising precursor for preparation of a bioengineered heparin using an enzymatic combinatorial approach, and generation of all kinds of substrates for the enzymes involved in the biosynthesis of heparin and heparan sulfate.
As a specific substrate, K5 polysaccharide can be degraded by K5 lyase A (Cat#: E-KFLA) into octa-, hexa-, tetra-, and disaccharide with Δ4,5GlcA at the non-reducing end which can be detected by UV spectroscopy at 232 nm. Meanwhile, it can also be used as a substrate for the N-deacetylase/N-sulfotransferase (NDST) enzyme.
